IN VITRO REGENERATION OF CASTOR PLANT UNDER THE INFLUENCE SUCROSE OSMOTIC STRESS

Title: IN VITRO REGENERATION OF CASTOR PLANT UNDER THE INFLUENCE SUCROSE OSMOTIC STRESS
Authors: OLUWOLE, LOLA VICTORIA
Abstract: Environmental stress results in water deficiency for plant, thus impairing its numerous
biological roles. In vitro screening for stress tolerance will have its significance in
identifying cultivars with optimal stress tolerance and productivity. Studies on castor plant
(Ricinuscommunis) could improve our understanding of the mechanism of plant resistance
to drought stress. This investigation was aimed at examining the effect of osmotic stress
generated by sucrose on shoot length, fresh weight shoot, fresh weight leaves, plant
regeneration and chlorophyll content related to drought stress. Nodes of three
Ricinusaccessions (cultivar 004, 005, 006) were cultured on MS (MS macro and
microelements) medium with addition of 1.6 mg BAP (6-benzylaminopurine) and 0.08 mg
of IAA (Indole acetic acid). Different levels of osmotic stress were induced by using two
concentration of sucrose (0% and 6%) in MS medium.
Accession 004 is resistance to drought stress than accession 005 and 006. Cultivar 004
under control and sucrose levels has the highest growth parameters such as shoot fresh
weight, plant regeneration percentage, plant height and leaves fresh weight than other
accessions 005 and 006 but there was no significant difference in leaves numbers among
the accessions. In turn cultivar 004 have the highest chlorophyll a and carotene than the
other accessions. Accession 006 showed reduction in chlorophyll a content and
physiological growth parameters than other accessions raised under 6% sucrose.
Therefore, accession 004 is resistance to drought stress whereas; accession 006 is a
drought sensitive. Farmers especially those in semi-arid and arid regions of Nigeria is
therefore encouraged to cultivate castor accession 004 in order to obtain optimal castor
yield.

Title: EFFECT OF DROUGHT STRESS TOLERANCE IN MAIZE maysL.)AND PEARL MILLET(PennisetumglaucumL.)
Authors: ALOZIE, PRECIOUS; Iwuala, E. N.
Abstract: This investigation is to determine the physiological basis of water deficit stress in pearl
millet,
each representing a treatment and replicated 3 times. Category
received 100ml of water every 2 days throughout the experimental period. Category
received 100ml of water every 2 days for 6 weeks before subjecting them to simulated
drought. Physiological parameters evaluated after the treatment include biomass, relative
water content of leaf, leaf area, chlorophyll level and leaf epidermal quantification. Analysis
of variance was also found to be significant for genotypes, treatments and their interactions at
0.05% level. The results showed drought caused decrease in biomass, chlorophyll, relative
water content
drought, while in Pennisetumglaucum, showed a significant increase in root/shoot ratio,
biomass, and abaxial epidermal surface under drought condition. Therefore, the comparative
analysis of these two genotypesunder well-watered and water deficit stress condition revealed
that P. glaucumwas superior over Z. mays indicating that such relationships can be positively
attributed to drought tolerance.

EFFECT OF SOILS IN SOME DIFFERENT LOCATIONS ON THE VEGETATIVE AND FLORAL CHARATERISTICS OF MAIZE ( UNIVERSITY OYE EKITI (FUOYE)MAIN CAMPUS

Title: EFFECT OF SOILS IN SOME DIFFERENT LOCATIONS ON THE VEGETATIVE AND FLORAL CHARATERISTICS OF MAIZE ( UNIVERSITY OYE EKITI (FUOYE)MAIN CAMPUS
Authors: ADEGBEYENI, HENRY OSEDIPO; AKINYELE, B.O.
Abstract: The effect of soils from five different locations of Federal University Oye Ekiti
the vegetative and floral characteristics of maize Boys Hostel, Theatre Art Complex, Old Hostel, New I.C.T Building and New Administrative Block. The experiment was a Completely Randomized Design the screen house of the department of Plant Science and Biotechnology. Two seeds were sown in each bucket and later thinned to one plant two weeks after planting. Watering was done at an interval of three days. The leaf length, leaf width, plant height, stem thickness,
number of leaves per plant and length of inflorescence were measured at the blooming stage using a tape graduated in centimeters. Data amassed were subjected to analysis of variance (ANOVA)and means separated using the Duncan’s Multiple Range Test (DMRT).Of all the vegetative attributes studied, only the soil obtained from the Theatre Art Complex
is significantly different
length, number of leaves per plant, plant height and they have higher leaf length, plant height and number of leaves per plant. Most importantly the length of inflorescence of the maizeplant of Theatre Art Complex is significant difference from other source of soil.
This study revealed that soil obtained from Theatre Art Complex is superior to other soil
source. This superiority may be due to the higher concentration of mineral nutrient require by maize plant which maybe least in others source of soil or may be due to the leaching of the soil. It may be due to the presence of Chromolaena odoratawhich is use as a fallow species and soil fertility improvement plant because Theatre Art Complex soil was surrounded by Chromolaena odorata
